When you see Diddy on the title screen Press and hold A, then press and hold B...You'll have 4 new characters. Also on the main title screen press A and B Simultaneously, You'll hewar Mario and Luigi say "Im the Best!" Press down on the vehicle selection and you'll have four new characters.
